What's the best way to go about it? I'd rather do it on one computer, for a group. Cheers.

D&D Insider is working on a Character Generator that would enable players to build their CHaracter quickly on the computer, and I'm guessing that leveling would be a quick and easy process too. It only covers low levels right now, but eventually it will be capable of going from level's 1 through 30, and incorproate ALL WotC content.

Also D&DI offers a compenduum that has all the "crunch", or mechanics so you have it all available without lugging a suitcase full of books. They are also working on a Dungeon builder utility.

I'm not sure if that's what you're looking for or not...

If you mean gaming online, there are some programs for that - basically a chat client + map + dice roller, usually. I've used openrpg a few times, it's passable. I used Screenmonkey once, and it is not bad either - but the person who wants to DM must purchase it.
